Microscopy data associated with manuscript: Cellular interactions in the sentinel lymph node predict melanoma recurrence

Description

In order to understand the immune-spatial environment of melanoma sentinel lymph nodes (SLN), this study profiled 43 tissues samples from a cohort Stage II melanoma that underwent SLN biopsy. All samples were randomly selected from archived samples between 2010-2017 at the University of Pennsylvania. The median age of the cohort was 61 and 76% were male.

  • 22 lacked tumor infiltration (SLN-)
  • 21 contained tumor micrometastases (SLN+)
  • 8 samples from Stage I SLN were also analyzed as a comparison, being the closet to a normal lymph node.

Formalin fixed paraffin embedded (FFPE) SLN from each patient were sectioned at 5 µm thickness. Tumor micrometastases within the SLN were identified by a melanoma pathologist using hematoxylin and eosin staining (H&E). 10 Stage II SLN that were SLN+ and 5 Stage I SLN were selected for multiplex immunofluorescence imaging using the COdetection by inDEXing platform (CODEX).  A high pH antigen retrieval buffer and photobleaching to reduce autofluorescence was incorporated into the CODEX experimental workflow.  Further information on image data generation, cleaning, and analysis is available in the cited paper.

Image data is separated into two categories:

  • Samples from Stage I SLN (15 total images)
  • Samples from Stage II SLN (30 total images)
